The Quota Nobody Reached
Why a trawler that stops fishing in March leaves most of its year's allowance in the sea — and why that arithmetic suggests that a medicinal chemistry programme's list of the properties that are hard to fix may be a record of the order in which its criteria were met, not of how hard any of them is.
Candidate · UnvalidatedI.The First Known Thing — the boat that stops in March
fisheries management · mixed fisheries and the choke
A demersal trawler in the North Sea does not catch one species. It catches what comes up in the net: cod, haddock, saithe, whiting, plaice, several of them in one tow, in proportions the skipper can shift by choosing grounds, gear and season but cannot separate at will. Management, however, is written stock by stock. Each stock has its own total allowable catch, and each vessel or producer organisation holds a share of each. Under the European Union's landing obligation, phased in between 2015 and 2019, the catch of a regulated stock may no longer be discarded at sea to keep fishing; it counts against the share. So the year ends, for that vessel, when the first of its shares runs out. The stock that runs out first is called the choke.
what the choke costs, in the arithmetic of the year
Mortensen and colleagues worked the sums for a single 28-metre Danish trawler in 2018 and found that its cod share, taken at the vessel's observed catch composition, would have been exhausted in late February or early April depending on the year — closing a fishery whose haddock, saithe and plaice shares were nowhere near spent. Karp and colleagues, reviewing quota use across mixed-stock fisheries in 2024, report that the average fraction of the allowable catch actually harvested runs between about a fifth and two thirds and has been falling, and that the losses fall on abundant stocks whose catch is limited by the concurrent catch of something else. The advisory machinery treats this as its central quantity. The mixed-fisheries framework used by the International Council for the Exploration of the Sea brackets a year between two scenarios: in one, each fleet stops when its last share is exhausted; in the other, each fleet stops when its first share is exhausted. The distance between those two numbers is not a property of any stock. It is a measurement of how badly the separate limits fit the mixture that the gear actually produces.
and the consequence the field has had to absorb
One consequence is administrative and is discussed constantly: which stock chokes depends on the fleet, the gear and the ground, so a limit that binds one vessel in February is never approached by another, and a fleet that redirects effort to avoid its choke changes the composition of its catch and moves the choke to somewhere else. A second consequence is quieter and is what matters here. Because fishing stops at the first limit, every other limit that year is never tested. The uptake recorded against a non-choking share — sixty per cent, thirty per cent — is not a statement about that stock's tightness at all. It is a statement about when the boat was stopped by something else. A manager reading a table of uptakes and concluding that haddock is not a constraining stock has read the order of a queue and called it a ranking of scarcity.
II.The Second Known Thing — the series and the property that killed it
drug discovery · lead optimisation and multi-parameter trade-offs
Walk to a hall where a chemical series is being improved. A lead series must satisfy many criteria at once — potency against the target, selectivity, solubility, permeability, metabolic stability, plasma protein binding, absence of a cardiac channel liability, absence of a genotoxic alert, a route and a dose that a patient could take. The chemistry that moves one of these moves the others; the parameters are produced jointly by the same molecule, in proportions a chemist can steer but cannot sever. The field's instruments for handling that are weighting instruments: desirability functions that map each property onto a common scale and combine them, efficiency indices that price potency against size or lipophilicity, and hard filters that refuse a compound outright on one axis.
how the record of failure is kept
Programmes keep a record of why series stopped. Attrition analyses count candidates lost and assign each loss to a cause — the analysis Waring and colleagues assembled in 2015 across four companies, covering several hundred candidates, is the best-known published instance — and within a project, attrition curves of the kind Munson and colleagues set out in the same year show how many of a series' compounds survive each successive criterion in the cascade. These tallies are what the field's shared beliefs about difficulty are made of. Everyone knows which properties kill programmes; the knowledge is largely the accumulated memory of which property was named on the death certificate.
and the structure of the cascade that produces the record
The record is produced by a screening cascade, and a cascade is an order. Cheap and early assays are run on everything; expensive, slow or animal assays are run only on what survived. A compound stopped on solubility is usually never put into a cardiovascular study; a series abandoned for metabolic instability does not go on to have its genotoxicity resolved. The order is not arbitrary — it is chosen for cost and for information — but it means the later assays are performed on a population that has already passed the earlier ones, and the properties at the end of the cascade are observed only on survivors. The field is aware of the effect in its everyday form: chemists say that a project's problems arrive one at a time, that fixing one liability uncovers the next, and that every series has a wall. What it has not done, so far as the published record shows, is treat the naming of the wall as a measurement that needs correcting before it can be read.
III.The Bridge
no quota paper cites a desirability function
The halls share nothing. Fisheries management and medicinal chemistry have no common literature, no shared instruments and no shared journals; where drug discovery has borrowed from outside it has borrowed from engineering optimisation, decision analysis and economics, and where fisheries science has borrowed it has borrowed from population dynamics and from the theory of common-pool resources. Nor is this the familiar observation that a system is limited by its bottleneck, which is a commonplace in both places and explains nothing by itself. What the two halls share is a specific structure: several outputs produced jointly by one effort, in proportions that can be steered but not separated, each output carrying its own independently set limit, and a stopping rule that fires at the first limit reached. One of the halls has been forced, by a change of law and an unforgiving arithmetic, to work out what such a structure does to its own records.
X: the limits that never bind are never tested
Call it choke censoring. Where one effort produces several outputs in proportions it can steer but not sever, each output is held under its own separately set limit, and the effort stops at the first limit reached, every other limit that occasion is left untested; the margin remaining against it records when the effort was stopped by something else, and not how tight it is. So a tally of which limits stopped the effort orders the limits by their position in a queue rather than by their severity, and because steering the effort changes the mixture and therefore which limit binds, the queue is itself the residue of past steering — the tally measures the history of the fleet, not the scarcity of the fish. Two things follow that are not the ordinary counsel to mind one's bottleneck. First, the correction is a known one: a limit that was never reached is a censored observation, and the severity of each limit is estimable from the occasions on which something else stopped the effort first, by the same arithmetic that recovers a survival curve from patients still alive at the end of a study — which means the field's ranking of its own difficulties is not merely uncertain but correctable, and correctable from records already kept. Second, the gap between stopping at the first limit and stopping at the last is a quantity of the mixture rather than of any component, and it is the thing that a scheme of separate limits cannot see: it can be computed, it has a unit, and nothing in a weighted score reports it.
IV.The Question That Becomes Possible
neither discipline would ask this alone
The chemistry hall counts the causes of death and reads the counts as a ranking of difficulty. The fisheries hall has no molecules and no assays, but it has had to learn, at the cost of tied-up boats, that an unspent allowance is not evidence of an easy stock. Neither asks whether a research field's shared sense of which properties are hard is an artefact of the order in which its criteria happen to be applied, nor whether the margin a dying series still held on everything that did not kill it is a measurable quantity worth keeping.
the choke-censoring hypothesis
Is the received ranking of which molecular properties are hard to fix — read off the tallies of what terminated compounds and series — a record of choke order rather than of difficulty, such that a property seldom named as terminal is not an easy property but an untested one, and does a censoring-corrected estimate of each property's severity, computed from programmes stopped by something else, reorder that ranking; and does a series' terminal choke gap — the margin still held against every criterion that did not stop it, at the moment the first one did — predict which series can be rescued when the choking property is later relaxed, solved, or removed by a change of criterion?
why it would matter
Because the ranking is load-bearing. It decides which predictive models a company builds and buys, which assays are moved earlier in the cascade, which structural alerts become hard filters, and which liabilities a portfolio refuses to start a programme against. If the ranking is partly an artefact of cascade order, then the properties the field treats as tractable are in part those its cascade never gets to, and effort has been allocated to the loudest rather than the largest. The choke gap matters for a nearer reason: it is a disposal decision. Series are shelved with their data, and when a choking criterion later moves — a new formulation option, a revised safety standard, a target's competitive landscape changing what potency is needed — someone must choose which shelf to reopen. A weighted score cannot help, because it collapses at the moment one axis fails. The margin held on everything else at the moment of failure is exactly what distinguishes a series that died of one thing from one that was dying of everything, and it is recoverable from records that already exist.
V.How It Could Be Proven — or Broken
a candidate must be breakable, or it is merely poetry
An intercognix candidate earns nothing until it names its own test. This one needs no new chemistry and no new assay. It needs a set of project histories with the property measurements attached and the stopping decision dated, which every large discovery organisation holds, and a partial public proxy in the deposited property panels and in the published series that record their own attrition.
What would support it
Take a decade of completed lead-optimisation projects within one organisation, where the cascade order, the criteria in force and the date of each stopping decision are recorded. For each series, mark which criterion it was stopped on, and for every other criterion record whether it had been measured, and if so, the margin held against it at that date — this is the choke gap, one number per criterion per series, with an explicit code for not measured. Then do three things. First, estimate each property's severity twice: once as the field does it, by the share of stopped series naming it, and once by treating a series stopped on another property as censored for this one, taking the hazard from the series that survived long enough to be tested. The claim is that the two rankings differ materially and in a particular direction — that properties sitting late in the cascade, and properties whose assays are expensive, rise when the censoring is corrected. If the two rankings agree, the whole construction is a correction to nothing. Second, test the endogeneity of the queue directly: within projects that performed a scaffold hop or changed series, ask whether the choke moved, and whether the property that bound before the hop is the one that binds after. The fisheries prediction is that it is not, and that the identity of the choke tracks the chemistry, so a chemist's belief about which property is hard should be predictable from what their last three series happened to be. Third, the rescue test, which is the part that could not have been guessed from either hall alone: assemble the cases where a choking criterion was later relaxed or solved — a criterion revised on new evidence, a formulation that took a solubility limit away, a backup programme restarting a shelved series — and ask whether the terminal choke gap, computed at the time of shelving and blind to what happened afterwards, separates the series that were successfully restarted from those that failed again immediately on a second property. A prospective version costs nothing: record the gap at every stopping decision from now on, and wait.
What would break it
It breaks first, and most likely, by collapsing into a measure already in use. The censoring-corrected ranking may simply reproduce the naive tally, which will happen wherever cascades are run in parallel rather than in sequence — many modern discovery organisations measure a full property panel on every compound, and where everything is measured on everything there is no censoring to correct, and the effect survives only at the level of the series decision, where the numbers are small. The choke gap may be worse: if it is strongly correlated with the minimum of the desirability scores the project already computes, or with the attrition curves of a lead-optimisation attrition analysis, then it is those quantities under a new name and the credit belongs where it sits. Second, the censoring may not be the ignorable kind. Assays are ordered by cost and by suspicion, so the properties measured late are measured late partly because the team already judged them unlikely to fail, and the decision to stop is a human one taken with all the data in view rather than a limit tripping mechanically; if the reason a property went unmeasured is itself informative about that property, the correction is not identified by any amount of arithmetic and the fisheries analogy has transferred a structure without transferring the independence that makes the structure estimable. Third, the properties may not be separable enough for a queue to exist. Lipophilicity moves solubility, metabolic stability, promiscuity and the cardiac channel liability together, so a series may not die of one thing at all; if the measured gaps on non-terminating properties are near zero in most cases, there is nothing to rank, everything reduces to a single axis the field already watches, and the interesting structure is absent from real data. Fourth, the record may not support the measurement. Stopping decisions are often undated, retrospectively attributed, or made for reasons — a competitor's publication, a portfolio review, a team dissolved — that have nothing to do with any property, and a tally contaminated by those is not a censored measurement of difficulty but a mixture with something that has no severity at all. Fifth, the analogy may mislead about what the limits are. A fishing quota is an administrative number set outside the vessel, identical for everyone holding a share and unchanged by anything the skipper does; a property criterion is set by the project, revised mid-programme, and different for an inhaled drug and an oral one — so the queue in chemistry may be so project-specific that no ranking across programmes exists to be corrected, which would make the whole exercise valid within a single project's history and empty across a field. Sixth, the rescue test may be unrunnable. Restarts of shelved series may simply be too rare to separate anything, in which case the hypothesis is not refuted but has no observations, and should be reported as such rather than as support.
the honest status
Note the narrowness. The claim is about one statistic computed from records that already exist, about the ranking of properties by difficulty within lead optimisation, and about one downstream event — the restart of a shelved series — for which ground truth exists inside companies and, thinly, in the published literature. It says nothing about how properties should be weighted, which is a different question with a large literature of its own, and it does not claim that any particular property is harder than believed; it claims only that the tally the belief rests on has a correctable bias whose direction is predictable from the cascade order. It is also worth saying what does not transfer. A fishery's limits are exogenous and its catch composition is measured for every species landed, whether or not it chokes; a chemistry programme measures a property only if it decided to, which is the one asymmetry that could sink the correction, and no argument from boats can settle it. What transfers is a way of reading a record — that an unspent margin against a limit is evidence about the stopping rule and not about the limit — and not the accounting that makes it clean at sea. The whole thing rests on a question that one afternoon with a company's project database would answer: whether the two rankings, naive and corrected, actually differ. If they do not, this is a correction to nothing, and should be said so plainly. Until that comparison is made, this remains exactly what its stamp says: a candidate. A structural echo between two halls of the library, waiting for someone with the tools of both to walk the corridor.
